Growth Conditions |
Temperature: 37°C
Recommendations for Infection: Plant cells 24-48 hours in advance and infect when cultures are 70-80% confluent. Remove medium, wash cell monolayer once with PBS, and inoculate with a small volume (eg. 1 mL per cm2) of virus diluted to provide a MOI of about 0.1. Adsorb 1-2 hours at 37°C in a humidified 5% CO2 atmosphere. End adsorption by adding virus growth medium.
Incubation: Incubate infected culture for 5-9 days at 37ºC in a humidified 5% CO2 atmosphere, until CPE are well advanced through 90% of the culture. |
