Restriction digests of the clone give the following sizes (kb): EcoRI--4.3, 1.7; HindIII--5.6, 0.5; BamHI--4.3, 1.2, 0.3; PstI--6.0. The insert contains the following sites (nucleotide position according to the published sequence): BamHI--514, 848; HindIII--1240, XbaI--1480. Crry and Cr-2 are within 10 kb of each other and are transcribed in the same direction. Encodes a single open reading frame starting at nt 482 of the published sequence and extending through the 3' end of the cDNA. Comparison with genomic sequence shows this clone lacks sequences for 7 additional amino acids. Spleen 5 cDNA (ATCC 63065) differs from liver 10 cDNA (ATCC 63064) by having an additional 129 nucleotides between the signal sequence and the nucleotides encoding the first 60 aa repeat. Spleen 5 also does not extend as far in the 5' direction. Liver 10 cDNA and spleen 5 cDNA are clones of alternative splice products of the Crry gene. The mRNAs are transcribed in all tissues examined in a 10 (liver 10) to 1 (spleen 5) ratio. When the 340 bp BamHI/BamHI fragment (lacking the extra 129 nucleotides) is used as a probe, the following EcoRI fragments are detected in BALB/c mice: 7.5 kb, 5.6 kb (chromosome 8), and 4.5 kb. Detects mRNA species of 1.9, 2.0, and 2.1 kb. |
