Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--2.686, 0.911; EcoRI/HindIII/SstI--2.686, 0.505, 0.406. Sonicated extracts of E. coli HB101 transformed with this construct express both ST and LTB immunoreactivity. The 0.406 kb EcoRI/SstI fragment encodes the heat stable enterotoxin, and the 0.505 kb SstI/HindIII fragment encodes the heat labile enterotoxin. The latter fragment, derived from EWD299 (ATCC 37218), includes the coding region for the B subunit and 3' transcription and translation stop signals, but lacks the ribosome binding site and sequences encoding the signal peptide. The former fragment is at the 5' end and includes the complete coding sequence and 5' regulatory regions (transcription initiation and ribosome binding site) but not the 3' transcription and translation stop signals. |
