Restriction digests of the clone give the following sizes (kb): EcoRI/KpnI--4.4, 1.4, 1.2; EcoRV--4.4, 2.6; EcoRI--4.4, 2.6. Expression from the aroF promoter results in an aroF-pheA fusion protein. Insert contains aa 1-351 of the aroF coding sequence and the complete pheA coding sequence. The construct contains a weak attenuator sequence at the 5' end of the pheA coding sequence. Production of phenylalanine is enhanced when the pheA structural gene is introduced as part of a synthetic operon, rather than as a single deregulated structural gene. |
